dc.description.abstract |
Lidský organismus je denně vystaven účinku volných radikálů. Ačkoli mohou být tyto molekuly
na jedné straně organismem využívány v jeho prospěch, na straně druhé ho mohou vážně poškodit. Nejvýznamnější volné radikály patří mezi reaktivní formy kyslíku a dusíku. Ochranu před oxidačním poškozením aktivními formami kyslíku zajišťuje řada obranných systémů. Antioxidační obranné mechanismy zahrnují neenzymové a enzymové systémy. Mezi nejvýznamnější enzymy řadíme glutathion-S-transferasu, glutathionreduktasu a glutathionperoxidasu. Glutathion-S-transferasa hraje důležitou roli při detoxikaci, kdy katalyzuje konjugaci hydrofobních a elektrofilních
látek s glutationem. Glutathionreduktasa je enzym, který zodpovídá za udržování poměru
koncentrací redukovaného a oxidovaného glutathionu uvnitř buněk. Glutath ionperoxidasa
katalyzuje redukci peroxidu vodíku na vodu a současně oxiduje glutathion. Aktivitu antioxidačních enzymů lze stanovit spektrofotometrickou metodou, jejímž principem je měření poklesu absorbance způsobené oxidací NADPH při 340 nm. Rychlost poklesu absorbance je přímo úměrná aktivitě daného enzymu. Mezi další metody vhodné pro stanovení aktivity těchto enzymů patří metody fluorimetrické, kolorimetrické či metody HPLC. |

The human organism is exposed daily to an effect of free radicals. Although these molecules can be used by the body in his favor, on the other hand they can damageit. Free radicals belong among reactive oxygen and nitrogen species. Protection against oxidative stress caused by reactive oxygen species is ensured by a number of defense systems. Antioxidant defense includes non-enzymatic and enzymatic compounds. The most important enzymes are glutathione-S-transferase,
glutathione peroxidase and glutathione reductase.
Glutathione-S-transferase plays an important role in detoxification. It catalyzes conjugation of hydrophobic and electrophilic compounds with glutathione. Glutathione reductase is a key enzyme for maintenance of high ratio of reduced glutathione and glutathione disulfide in cells.
Glutathione peroxidase catalyzes reduction of hydrogen peroxide to water together with oxidation of glutathione. The activity of antioxidant enzymes can be determined by spectrophotometric methods. These metods are mostly based on the measurement of absorbance decline due to oxidation of NADPH (λ =340 nm). The
decline in absorbance is directly proportional to the enzyme activity. Other methods for determination of enzyme activity are fluorometric methods, colorimetric and HPLC methods. |
